مقدمه: نقش آسیکلویر (ACV)در ایجاد سمیت و مرگ سلولی در دستگاه تولیدمثل شناخته شده است. آسکوربیک اسید (VC) و منتون (M) دو آنتیاکسیدان ضروری در نقش حفاظت برای ملکولی زیستی مهم شناخته شده است.
هدف: تعیین تأثیر نقش حفاظتکنندگی آنتیاکسیدان VC و M در برابر آسیب DNA وارده توسط آسیکلویر (ACV) در اسپرماتوزوآی موش صحرایی نر.
موارد و روش ها: در این مطالعه تجربی موش صحرایی نر نژاد ویستار با متوسط وزنی10±250 گرم به شش گروه تقسیم شدند (تعداد هر گروه 18) mg/kg/day) 15)ACV mg/kg/day),20 mg/kg/d) ,ACV+Vit C (100µlm/d) ,ACV+ menthone (250,ACV+ menthone( µlm/day)400) ACV+ menthone و گروه شاهد بدون هیچ تیماری تقسیم شدند. در پایان 48 روز تحت رژیمهای غذایی خاص بافت اپیدیدیم هر موش صحرایی خارج و اسپرمهای اپیدیدیمی در محلول PBS استخراج گردید. جهت بررسی با رنگ آمیزی تانل از طریق فلوروسنت میکروسکوپ درصد مثبت اسپرماتوژوآی با رنگ آمیزی تانل ارزیابی شد. هر نمونه برای ارزیابی سه بار تکرار گردید.
نتایج: موشهای نر تحت درمان ACV در مقایسه با گروهای دیگر در آسیب DNA تفاوت معنیدار داشت. در صد مثبت اسپرماتوژوآی با رنگآمیزی تانل 90/83 (0/001p<) در گروه ACV بیانگر قدرت باروری پائین در موش نر بود. VC و نیز M در بالاترین دوز که بیانگر قابلیت آنتی اکسیدان و عوامل ضدالتهابی در جلوگیری از اثر آپوپتوز آسیکلویر بر روی DNA اسپرمها نشان دادند. نتایج در گروه VC در جلوگیری از اثر آپوپتوز آسیکلویر بر روی DNA اسپرم 16/38 (0/001p<) و در گروه منتون با دوز بالا همچنین پایین نشان داد 16/05 (0/001p<).
نتیجه گیری: نتایج نشان داد که آسکوربیک اسید و منتون نقش حمایت کنندگی بسیار قوی در بالاترین دوز باعث جلوگیری از اثر آپوپتوز آسیکلویر بر روی DNA اسپرمها و داستگاه تولیدمثل موش صحرایی نر دیده شد.
