مطالعه مقایسه‌ای نانوذرات نیوزومی حاوی عصاره‌های درمنه کوهی و درمنه دشتی بر تکثیر سلول‌های سرطانی پستان رده MCF‑7: یک مطالعه آزمایشگاهی. International Journal of Reproductive BioMedicine. 1405; 24 (8) :721-738

مقدمه: سرطان پستان دومین عامل مرگ­و­میر ناشی از سرطان در میان زنان است و سالانه تقریباً 30% از کل تشخیص‌های جدید سرطان در زنان را به خود اختصاص می‌دهد.
هدف: با توجه به شیوع سرطان پستان در زنان، هدف اصلی این مطالعه توسعه یک سیستم دارورسانی مؤثر برای درمان این بیماری با استفاده از نیوزوم‌های حاوی گیاهان درمنه است.
مواد و روش ­ها: این مطالعه آزمایشگاهی شامل استخراج هیدروالکلی درمنه با استفاده از 2 روش خیساندن و سوکسله و به دنبال آن ارزیابی خواص آنتی‌اکسیدانی بود. نانوذرات نیوزومی با استفاده از روش هیدراتاسیون لایه نازک سنتز شدند. مشخصه‌یابی نانوذرات، شامل اندازه ذرات و پتانسیل زتا، با استفاده از پراکندگی نور دینامیک (Zeta Sizer) و طیف‌سنجی فروسرخ تبدیل فوریه (FTIR) انجام شد. ارزیابی سمیت سلولی با استفاده از سنجش MTT انجام و تجزیه و تحلیل آماری با استفاده از ANOVA یک‌طرفه و تعیین معناداری (p-value) صورت گرفت.
نتایج: این مطالعه نشان داد که عصاره درمنه کوهی، به ویژه زمانی که با روش سوکسله و اتانول 60% به دست آید، فعالیت آنتی‌اکسیدانی برتر و محتوای فنولی/فلاونوئیدی بالاتری نسبت به درمنه دشتی نشان می‌دهد. نانوذرات نیوزومی بهینه­شده بازده کپسوله‌سازی 87% برای درمنه کوهی (F3) و 93% برای درمنه دشتی (F6) نشان دادند. میزان رهایش تجمعی در مدت ۷ ساعت برای عصاره گیاه درمنه دشتی 71% (F3) و برای عصاره درمنه کوهی 73% (F4) بود. نانوذرات بهینه‌شده اندازه‌های ۱۱۵ نانومتر برای عصاره درمنه کوهی (F6) و ۷۹ نانومتر برای درمنه دشتی (F3) را همراه با پتانسیل زتا به ترتیب 6/20- میلی‌ولت و 2/25- میلی‌ولت نشان دادند. آنالیز FTIR بارگذاری موفق و برهمکنش‌های بین‌مولکولی مطلوب را تأیید کرد. سمیت سلولی نانوذرات کپسوله شده در برابر سلول‌های MCF-7 افزایش معناداری نسبت به فرم آزاد نشان داد (05/0 > p)، در حالی‏که نیوزوم‌های خالی سمیت قابل توجهی نداشتند. مقادیر IC₅₀ برای درمنه کوهی کپسوله شده 4/254 میکروگرم بر میلی‌لیتر و برای درمنه دشتی کپسوله شده 8/405 میکروگرم بر میلی‌لیتر بود.
نتیجه ­گیری: بر اساس نتایج به‌دست‌آمده، استفاده از عصاره‌های گیاهی و نانوذرات نیوزومی می‌تواند گامی مثبت در جهت درمان سرطان پستان محسوب شود.
