fa اثر سن بر بعضی از نشانگرهای آپوپتوز و استرس اکسیداتیو در مردان نابارور نورمواسپرم و الیگواسپرم: یک مطالعه مقطعی Effect of senescence on some apoptosis and oxidative stress markers in infertile normozospermic and oligospermic men: A cross-sectional study Reproductive Genetics Original Article Original Article مقدمه: افزایش سن در مردان ممکن است که بر روی عملکرد بیضهها و پارامترهای اسپرم و تولید سطح بالای اکسیدانها و آپوپتوز اثر داشته باشد. هدف: در این مطالعه اثرات سن مردان بر روی بیان بعضی از نشانگرهای آپوپتوز و استرس اکسیداتیو در مایع سیمن در یک مرکز بهداشت نیجریه بررسی شد. موارد و روشها: در این مطالعه مقطعی 122 مرد با سن 20 تا 60 سال که به جهت ناباروری تحت درمان بودند به حسب سن به چهار گروه تقسیم شدند. کاسپاز 3 سمینال، سیتوکروم C و ظرفیت کلی آنتیاکسیدان (TAC) بوسیله تکنیک الیزا بررسی و آنالیز دستی سیمن تحت استانداردهای WHO انجام گردید. نتایج: کاسپاز 3 سمینال و فعالیت سیتوکروم C با افزایش سن افزایش یافت، در حالیکه TAC و نشانگرهای اسپرم کاهش یافتند. سیتوکروم C                 (0/288r=، 0/002p=) و کاسپاز 3 (0/250r=، 0/005p=) به طور مشخص در ارتباط با سن در مردان نورمواسپرم بود در حالیکه سیتوکروم C (0/314r=، 0/02p=)، کاسپاز 3 (0/268r=، 0/05p=)، TAC (0/342-r=، 0/01p=)، و درصد مرفولوژی (0/414-r=، 0/002p=) در ارتباط با سن در مردان نابارور الیگواسپرم بود. نتیجه گیری: نشانگرهای اندازهگیری شده آپوپتوز با افزایش سن در مردان نابارور افزایش یافت، درحالیکه TAC و نشانگرهای آپوپتوز و استرس اکسیداتیو در ارتباط با سن مردان نورمواسپرم بود، ولی اثر سن بر روی نشانگرهای اسپرم در میان مردان اولیگواسپرم شدیدتر بود. بنابراین این نشانگرها میتوانند در مردان مسن مراجعه کننده به کلینیکهای ناباروری مورد توجه قرار گیرند. Background: Male senescence may affect testicular function, sperm indices and generation of high levels of oxidants and apoptosis. Objective: This study evaluates the effect of male age on the expression of some apoptosis and oxidative stress markers in seminal fluid of males investigated for infertility in a tertiary health institution in Nigeria. Materials and Methods: In this cross-sectional study, 122 men aged 20-60 yr who were investigated for infertility and were stratified according to age into four groups. Seminal plasma caspase 3, cytochrome C, and total antioxidant capacity (TAC) were assayed by ELISA technique, while manual semen analysis was performed according to WHO standard. Results: Seminal caspase 3, and cytochrome C activity increased while TAC and sperm indices decreased with increasing age. Cytochrome C (r=0.288; p=0.002) and caspase 3 (r=0.250; p=0.05) correlated significantly with age in normospermia while cytochrome C (r=0.314; p=0.02), caspase 3 (r=0.268; p=0.05), TAC (r=-0.342; p=0.01) and morphology percentage (r=-0.414; p=0.002) correlated with age in oligospermic infertile males. Conclusion: The measured apoptotic markers increased with increasing age while TAC and sperm indices decreased with increasing age of subjects evaluated. Although the levels of measured apoptosis and oxidative stress markers correlated with age in normozospermia, the effect on sperm indices was severe among oligospermia compare to normozospermia.
