en تشخیص حذف 4977-bp از DNA میتوکندری در زنان مبتلا به شکست در لقاح داخل آزمایشگاهی مطالعه مورد-شاهدی Detection of 4977-bp deletion of mitochondrial DNA in in vitro fertilization failure women: A case-control study Original Article Original Article مقدمه: کیفیت تخمک اغلب به عنوان یک عامل محدودکننده باروری، به خصوص IVF، در نظر گرفته میشود. برخی از جهشهای میتوکندریایی، به ویژه حذف 4977 جفت بازی، با افزایش سن، افزایش مییابد. بنابراین، این جهش میتواند به عنوان یک نشانگر برای پیری سلول در نظر گرفته شود، که کاهش کیفیت تخمکها را برای لقاح نشان میدهد. در برخی مطالعات پیشنهاد شده است که این (جهش) میتواند در سلولهای خونی زنان مبتلا به شکست  IVF مورد بررسی قرار گیرد. هدف: 1-تعیین فراوانی حذف 4977 جفت بازی در زنان مبتلا به شکست  IVFو 2-بررسی ارتباط بین حذف 4977 جفت بازی و سن بیماران. موارد و روش ها: واکنش زنجیرهای پلیمریزاسیون (PCR) برای تشخیص حذف 4977 جفت بازی در نمونههای خون 52 بیمار مبتلا به شکستIVF  و 52 زن سالم که حداقل یک فرزند سالم داشتند، استفاده شد. نتایج: براساس نتایج بدست آمده، 07/48% از زنان مبتلا به شکست IVF و 62/32 درصد زنان سالم دارای حذف 4977 جفت بازی میتوکندریایی بودند (0/163p= و 1/749 OR:). همچنین، در رابطه با سن این بیماران و فراوانی جهش 4977 جفت بازی، P-value و OR به ترتیب 0/136 و 1/749 بدست آمد. نتیجه گیری: براساس یافته های این مطالعه، رابطه معناداری بین فراوانی جهشهای 4977 جفت بازی میتوکندریایی و شکست در IVF وجود نداشت. Background: The quality of oocyte is often considered as a limiting factor for fertility, especially IVF. Some mitochondrial mutations, particularly the 4977-bp deletion increase with the age. Thus, this mutation can serve as a marker for cell aging, which indicates the reduced quality of the oocytes for fertilization. It has been suggested that this can also be investigated in the blood cells of women with IVF failure. Objective: 1-Determination of the frequency of 4977-bp deletion in women with IVF failure, 2-Investigation of the relationship between 4977-bp deletion and the age of patients. Materials and Methods: Polymerase chain reaction was used to detect the 4977-bp deletion in blood samples of 52 IVF failure women and 52 women who had at least one healthy child. After polymerase chain reaction with deleted and wild-type primers, the products were examined using agarose gel electrophoresis. Results: 48.07% of women with IVF failure and 34.62% of healthy women had a mitochondrial 4977-bp deletion, with p=0.163 and OR: 1.749. Also, in association with the age of these patients and the frequency of 4977-bp mutation, p and OR were obtained 0.163 and 1.749, respectively and frequency of this mutation was higher in patients over 35 yr old compared to other subgroups (Patients ≥35: 57.69).
