en ارزیابی پارامترهای اسپرم، استرس اکسیداتیو و اثرات هیستوپاتولوژیک ویتامین B12 در پیشگیری از سمیت بیضه ناشی از هلیکوباکتر پیلوری: یک مطالعه تجربی Evaluation of the sperm parameters, oxidative stress, and histopathological effects of vitamin B12 in preventing Helicobacter pylori-induced testicular toxicity: An experimental study Reproductive Epidemiology Original Article Original Article <div style="text-align: justify;">مقدمه: هلیکوباکتر پیلوری (H. pylori) با تأثیر بر سیستم تولیدمثل باعث عقیمی میشود. ویتامین B12 کمیت و عملکرد اسپرم را بهبود میبخشد. هدف: حفاظت ویتامین B12 در برابر اثرات نامطلوب H. pylori مورد بررسی قرار گرفت. مواد و روش ها: 40 سر موش نر C57 (6 هفتهای) به طور تصادفی به 4 گروه مساوی (10 =n ) شامل گروه 1 (کنترل بدون مداخله)، گروه 2 (H)، گروه 3 (HP) و گروه 4 (HB) تقسیم شدند. سه گروه تیمار به ترتیب 109×1 واحد تشکیل کلنی (CFU) H. pylori، CFU‌ 109×1 از H. pylori + فسفات سالین بافر (PBS)‌ و CFU 109×1 از H. pylori +µg/kg  50 از ویتامین B12 به ترتیب به صورت داخل صفاقی دریافت کردند. در گروههای القایی، H. pylori   3 بار با 1 سیسی PBS در طول روز به صورت دهانی تزریق شد. سپس پارامترهای مورفولوژی بیضه، تحرک، زندهمانی، تعداد و شکل اسپرم، سطوح پلاسمایی مالوندیآلدئید (MDA)، سوپراکسید دیسموتاز (SOD)، گلوتاتیون پراکسیداز (GPX) و ظرفیت آنتیاکسیدانی کل (TAC) اندازهگیری شد. همچنین تغییرات بافت بیضه با استفاده از نمرات جانسون، شاخص تمایز لولهای (TDI) و شاخص اسپرماتوژنز (SPI) مورد بررسی قرار گرفت. سطح سرمی ویتامین B12، هموسیستئین و تستوسترون نیز بررسی گردید. نتایج: نتایج نشاندهنده کاهش معنیدار نمره جانسون، TDI، SPI و سطح سرمی تستوسترون و هموسیستئین و همچنین افزایش معنیدار سطح MDA در گروه H و HP نسبت به گروه HB بود (05/0 p <). در مقابل، بیشترین فعالیت آنزیم‌های SOD و GPX و TAC و همچنین کمترین سطح سرمی MDA در گروه HB نسبت به سایر گروه‌ها مشاهده شد (05/0 p <). نتیجه گیری: ویتامین B12 منجر به افزایش فعالیت آنزیم آنتیاکسیدانی و پارامترهای اسپرم و همچنین کاهش آسیب به بافت بیضه شد. میتوان از آن به عنوان یک آنتیاکسیدان قوی در کاهش آسیب بیضه ناشی از H. pylori استفاده کرد.</div> <div style="text-align: justify;">Background: Helicobacter pylori (H. pylori) causes sterility by affecting the reproductive system. Vitamin B12 improves sperm quantity and function. Objective: Vitamin B12 protection against H. pylori adverse effects was investigated. Materials and Methods: 40 C57 male mice (6 wk) were randomly assigned to 4 equal groups (n = 10) including, group 1 (control without any intervention), group 2 (H), 3 (HP), and 4 (HB) received 1×109 colony forming unit (CFU) of H. pylori, 1×109 CFU of H. pylori+phosphate buffered saline, 1×109 CFU of H. pylori+50 μg/kg vitamin B12 intraperitoneally, respectively. In the induction groups, the H. pylori was orogasterically injected 3 times with 1 cc phosphate buffered saline throughout the day. Then testicular metrics, sperm motility, viability, quantity, and shape, plasma levels of malondialdehyde (MDA), superoxide dismutase, glutathione peroxidas, and total antioxidant capacity were measured. Also, testicular-tissue changes were examined using Johnson scores, tubular differentiation index, and spermatogenesis index. Vitamin B12, homocysteine, and testosterone serum levels were examined. Results: The results showed a significantly lower Johnson score, tubular differentiation index, and spermatogenesis index, and serum level of testosterone and homocysteine as well as a higher MDA level in the H and HP groups than the HB group (p < 0.05). In contrast, the highest superoxide dismutase and glutathione peroxidase enzymes activity and total antioxidant capacity as well as the lowest serum level of MDA were found in the HB group compared to other groups (p < 0.05). Conclusion: Vitamin B12 increased antioxidant enzyme activity, enhanced sperm parameters, and decreased injury to testicular tissue. It can be used as a potent antioxidant in reducing testicular damage induced by H. pylori.</div> : هلیکوباکتر پیلوری, پزشکی تولید‌مثل, استرس اکسیداتیو.
