en تغییر میزان بیان microRNA در بیماران الیگواسپرمیک Altered of microRNA expression level in oligospermic patients Original Article Original Article مقدمه: microRNA (miRNA) رشته RNA تک زنجیرهای کوچک با منشا داخلی است که از طریق مکانیسمهای مختلف بیان ژن را در مرحله پس از ترجمه تنظیم میکند، از این طریق اعمال اصلی سلولی از جمله تمایز، تکثیر، تکامل و آپوپتوز را در سلولهای جنسی تحت تأثیر قرار میدهد. هر گونه جهش و یا انحراف در بیان آنها میتواند منجر به اختلالات متعدد از جمله ناباروری گردد. هدف: هدف از این تحقیق بررسی سطح بیان hsa-mir-21،has-mir-22  و گیرنده استروژن بتا (ERβ) در بیماران نابارور الیگو اسپرمیک و مردان بارور کنترل همچنین ارتباط آنها با همدیگر میباشد. مواد و روشها: در این تحقیق تغییر در سطح بیان mir-22، mir-21 و ژن هدف مشترک آنها یعنی ERβ در مردان نابارور الیگواسپرمیک (43 نفر) در مقایسه با گروه کنترل سالم بارور با استفاده از روش Real-Time PCR مورد ارزیابی قرار گرفت. نتایج: آنالیز بیان mir-21،mir-22  با استفاده از متد qRT-PCR نشان داد که سطح بیان آنها به طور معنادار در افراد نابارور نسبت به گروه کنترل بالاتر بود (0/0001>p) و نیز سطح بیان ERβ در مقایسه با گروه کنترل پائین تر بود (0/0001>p). نتیجهگیری: نتایج ما نشان داد که mir-21، mir-22 میتوانند به طور غیر مستقیم از طریق تنظیم بیان گیرنده استروژن در روند اسپرماتوژنز دخیل باشند و ممکن است دارای ارزش تشخیصی و پیش آگهی در ناباروری داشته باشند. Background: MicroRNA (miRNA) is small endogenous, single strand RNA molecules that regulate gene expression at post-transcriptional level through several mechanisms to affect key cellular event including male germ cells differentiation, proliferation, development and apoptosis. Mutation and/or aberrant expression of miRNAs have been associated with progression of various disorders, including infertility. Objective: The purpose of this research was to study the estrogen receptor beta (ERβ), hsa-mir-21 and, hsa-mir-22 expression level in oligospermic infertile and control fertile men and correlation between them. Materials and Methods: In this study, the change in mir-21, mir-22 expression and their common target gene (ERβ) expression levels were evaluated in oligospermic infertile men (n= 43) compared with 43 age matched healthy control by Real-Time PCR methods. Results: Expression analysis by qRT-PCR test on miRNA have identified that mir-21, mir-22 levels were significantly higher than those in normal controls (p<0.0001) and ERβ expression level significantly decreased in comparison with the normal group (p<0.0001).
