مقدمه: افزایش دمای بیضه می­تواند منجر به آسیب اسپرماتوژنز و در نتیجه ناباروری مردان شود. سلول­های زایا که حساس به حرارت هستند، یکپارچگی ژنتیکی خود را از طریق مکانیزم محافظت از خود سلول یا از طریق مرگ سلول حفظ می­کنند. با این حال، تمایز کیفی در مورد اینکه چگونه گونه­های فعال اکسیژن و آنزیم­های آنتی­اکسیدان تنظیم مسیرهای سیگنالینگ آسیب سلولی، از جمله تجزیه DNA در دماهای مختلف را تنظیم می­کنند هنوز مورد بررسی قرار نگرفته است.
هدف: این مطالعه به منظور تعیین تأثیر استرس اکسیداتیو وابسته به گرما بر روی سلول­های زایای جنس نر انجام شد. همچنین، تغییرات کیفی وابسته به زمان در مرگ سلول­های زایا مورد مطالعه قرار گرفت.
موارد و روش­ ها: تعداد 30 سر موش صحرایی نر نژاد ویستار به صورت تصادفی به 5 گروه تقسیم شدند (6=n در هر گروه) ، یعنی کنترل، 30، 45، 60 و 90 دقیقه با توجه به پروتکل درمان حرارتی. مطالعات کمی DNA و DNA نردبان همراه با پارامترهای بیوشیمیایی مختلفی نظیر پراکسیداسیون لیپید (LPO)، گلوتاتیون، کاتالاز، سوپراکسید دیسموتاز (SOD)، گلوتاتیون پراکسیداز (GSH-Px)، گلوتاتیو ردوکتاز (GR)، گلوتاتیون s ترانسفراز (GST) انجام شد.
نتایج: حیواناتی که تحت درمان گرمایی 30 دقیقه و 45 دقیقه قرار گرفتند، واکنش سیستمی و تدریجی به استرس گرمایی را نشان دادند. در حالی که حیوانات تحت درمان 60 دقیقه و 90 دقیقه، یک تغییر معمول و ناگهانی محیط داخلی سلول­های زایا را نشان دادند. به ترتیب آسیب DNA laddering و smearing در 30 و 45 دقیقه و 60 و 90 دقیقه تحت درمان با گرما دیده شد.
نتیجه­ گیری: با افزایش طول مدت درمان حرارت، میزان آپوپتوز به سطح مطلوبی رسیده و افزایش طول در معرض قرارگیری حرارت، حالت مرگ سلولی از آپوپتوز به نکروز را به طور ضمنی به علت حمله شدید اکسیداتیو تبدیل می­کند.
