تأثیر غلظت‌های مختلف پلاسمای غنی از پلاکت بر ژن‌های مسیر آپوپتوز و تمایز سلول‌های بنیادی در نارسایی تخمدان ناشی از سیکلوفسفامید در مدل موشی: یک مطالعه تجربی. International Journal of Reproductive BioMedicine. 1404; 23 (8) :637-646

مقدمه: پلاسمای غنی از پلاکت (PRP)، به عنوان یک روش درمانی جدید، پتانسیل بازسازی بافت تخمدان از کار افتاده و کاهش علائم بیماران مبتلا به نارسایی زودرس تخمدان (POF) را دارد.
هدف: این مطالعه به بررسی تأثیر غلظت‌های مختلف پلاکت بر عملکرد تخمدان، بررسی ژن‌های آپوپتوز لنفوم سلول B 2 (BCL-2) و پروتئین X مرتبط با BCL-2 (BAX) و ژن تمایز فاکتور رونویسی متصل شونده به اکتامر 4 (OCT4) پرداخت.
مواد و روش­ ها: در این مطالعه، 30 موش سوری ماده (10-8 هفته، 30-25 گرم) به طور تصادفی به 5 گروه (6 = n در هر گروه) تقسیم شدند: 1- کنترل نرمال، 2- POF + PBS، 3- ml/kg 25/0POF + PRP ، 4- ml/kg 5/0 POF + PRP، 5- ml/kg 1 POF + PRP. از داروی           سیکلوفسفامید (CTX) برای ایجاد مدل موشی POF استفاده شد. پس از تزریق PRP و PBS، 2 هفته به موش­ها زمان داده شد. سپس موش­ها تحت عمل جراحی قرار گرفتند و تخمدان­ها برای انجام کارهای بافت­شناسی و مولکولی خارج شدند.
نتایج: در بررسی بافت­شناسی، توزیع انواع مختلف فولیکول در گروه POF + PRP همانند گروه کنترل بود. بیان ژن BAX در گروه­هایی که PRP دریافت کردند به طور معنی­داری کاهش یافت (001/0 > p). بیان ژن‌های BCL-2 و OCT-4 نیز در گروه‌های PRP افزایش یافت (05/0> p)، اما بیان ژن در غلظت‌های مختلف پلاکتی متفاوت بود.
نتیجه­ گیری: نتایج نشان داد که PRP می­تواند در POF مؤثر باشد، اما دوزهای مختلف PRP می­تواند اثرات متفاوتی بر بهبودی تخمدان داشته باشد.
